The small plane crashed into a hangar at Parafield Airport in northern Adelaide at about 2.10pm local time (5.40pm GMT).
The airport has been closed to all air traffic.
It is unknown how many people were on board the plane.
An eyewitness reported seeing black smoke billowing from a massive fire at the airport.
Multiple fire crews are in attendance and local residents have been urged to avoid the area.
Due to the fire, local people have also been asked to close their windows and doors.
